Tamoxifen dihydrogen citrate
Indications/Uses
Adjuvant therapy following primary treatment of mammary carcinoma & metastatic mammary carcinoma.
Dosage/Direction for Use
20-40 mg daily. Duration: 5 yr in adjuvant treatment of early hormone receptor-+ve mammary carcinoma.
Administration
Should be taken with food: Take w/ some liqd, do not chew.
Contraindications
Hypersensitivity. Women w/ history of DVT or pulmonary embolus & who require concomitant coumarin-type anticoagulant therapy. Pregnancy & lactation. Childn.
Special Precautions
Severe thrombopenia, leukopenia or hypercalcaemia. Increased risk of endometrial malignancies; causes of vag bleeding in post-menopause & irregular bleeding in pre-menopause should be clarified. Perform annual gynecological exam for non-hysterectomized patients; ophth exam prior to therapy.
Adverse Reactions
Hot flushes, ostealgia, pain in the affected tissue, transient anaemia, leukopenia, thrombocytopenia, hypercalcaemia, fluid retention, somnolence, headache, depressive mood, dysopia, thromboembolism, inter alia deep venous thromboses, pulmonary embolism, nausea & vomiting, hypersensitivity reactions, vulval pruritus, vag bleeding, uterine myoma, ovarian cysts, endometrial hyperplasia, endometrial polyps & endometrial malignancies.
Drug Interactions
Hormonal prep containing oestrogen, thrombocyte-aggregation inhibitors, anticoagulants, aminoglutethimide.
